新疆芜菁果胶多糖组成分析及免疫活性评价

Composition Analysis and Immunoactivity Evaluation of Pectic Polysaccharide from Brassica rapa L. in Xinjiang Uygur Autonomous Region

  • 摘要: 为分离纯化新疆芜菁果胶多糖,并对其进行组成分析及免疫活性评价。本论文采用水提醇沉法结合不同凝胶柱色谱制备新疆芜菁果胶多糖,理化分析表明:该果胶多糖纯品的总糖含量为78.86%±1.01%、糖醛酸的含量为15.82%±1.05%、硫酸基的含量为0.14%±0.05%,但不含有蛋白质和总酚;经傅立叶红外光谱分析,新疆芜菁果胶多糖纯品为同时含有α-及β-构型糖苷键的吡喃糖;HPGPC测得分子量为838 kDa;PMP衍生化分析发现由鼠李糖与半乳糖醛酸两种单糖组成,其摩尔比为1.94:98.06;通过RAW264.7细胞模型,发现当新疆芜菁果胶多糖浓度在12.5~100 μg/mL时,能显著提高细胞增殖活性与吞噬能力,有效刺激细胞释放NO、TNF-α、IL-1β和IFN-γ细胞因子,且呈现明显的剂量效应,说明新疆芜菁果胶多糖具有较强的免疫调节活性,可以作为天然免疫调节剂应用于功能食品或者制药工业。

     

    Abstract: In order to isolate and purify pectic polysaccharide from Brassica rapa L.(BRP)in Xinjiang Uygur Autonomous Region,and study its composition and immunoactivity evaluation. In this paper,BRP was prepared by water extraction and alcohol precipitation and combined with different gel column chromatography. Physicochemical analysis showed that the carbohydrate content of the BRP was 78.86%±1.01%,the uronic acid content was 15.82%±1.05%,and the sulfuric radical content was 0.14%±0.05%,while the protein and total polyphenols were not detected. Fourier transform infrared spectroscopy(FT-IR)indicated that the BRP was pyranose containing both α- and β-configuration in the sugar units. The high performance gel permeation chromatography(HPGPC)analysis showed that the molecular weights of the BRP was calculated to be 838 kDa. PMP derivatives analysis indicated that the BRP was composed of rhamnose and galacturonic acid in a molar ratio of 1.94:98.06.The RAW264.7 cells model found that,when the concentration of the BRP was 12.5~100 μg/mL,it could significantly improve cell proliferation activity and phagocytic ability,and effectively stimulate the release of cytokines such as NO,TNF-α,IL-1β and IFN-γ. Furthermore,an immunoactivity of the BRP was concentration dependent. It indicated that the pectic of Brassica rapa L. in xinjiang Uygur Autonomocs Region had strong immunoactivity. Hence,it could be used as a natural immunomodulator in functional foods or medicines.
