五味子多糖对对乙酰氨基酚诱导的小鼠肝细胞凋亡的影响

Effect of Schisandra chinensis Polysaccharide on Apoptosis of Hepatocytes Induced by Acetaminophen in Mice

  • 摘要: 目的:研究五味子多糖(Schisandra chinensis polysaccharide,SCP)对对乙酰氨基酚(Acetaminophen,APAP)诱导小鼠急性肝损伤的保护作用,并探讨其对肝细胞凋亡的影响。方法:采用一次性腹腔注射APAP(250 mg/kg)建立小鼠急性肝损伤模型,将50只ICR小鼠随机分为对照组、肝损伤模型组(MOD)和25、50、100 mg/kg SCP组。检测小鼠血清中谷丙转移酶(ALT)、谷草转氨酶(AST)与肝组织中谷胱甘肽(GSH)及丙二醛(MDA)水平;HE染色观察肝脏病理学变化,Hoechst 33258染色观察肝细胞凋亡情况;Western blot法检测肝组织中凋亡相关蛋白JNK、Bax、Bcl-2、Cleaved caspase-3的表达情况。结果:与MOD组相比,25及50 mg/kg SCP组小鼠血清中ALT、AST水平显著降低(p<0.05),100 mg/kg SCP组ALT、AST水平极显著降低(p<0.01);50 及100 mg/kg SCP组肝组织中GSH水平显著升高、MDA水平显著降低(p<0.05);SCP各组均不同程度改善肝细胞变性坏死及炎性浸润,并抑制肝细胞凋亡。同时,与MOD组相比,SCP预处理p-JNK蛋白表达显著降低(p<0.05),Bax、Cleaved caspase-3极显著降低,Bcl-2蛋白表达极显著升高(p<0.01)。结论:SCP对APAP诱导小鼠肝损伤具有保护作用,且其机制可能与抑制肝细胞凋亡有关。

     

    Abstract: Objective:To observe the protective effect of Schisandra chinensis polysaccharide(SCP)on acute liver injury induced by acetaminophen(APAP)in mice and explore its effect on hepatocyte apoptosis. Methods:The acute liver injury model in mice was established by intraperitoneal injection of APAP(250 mg/kg). 50 ICR mice were randomly divided into control group,model group(MOD)and 25,50 and 100 mg/kg SCP group. The levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum of mice,the levels of glutathione(GSH)and malondialdehyde(MDA)in liver homogenate of mice significantly measured. HE and Hoechst 33258 staining were performed for observing the change of hepatic pathology and apoptosis of hepatocyte,respectively. The expressions of apoptosis-related proteins JNK,Bax,Bcl-2 and Cleaved caspase-3 in liver tissue were detected by Western blotting analysis. Results:Compared with the MOD group,the levels of ALT and AST in serum of mice decreased(p<0.05)in 25 and 50 mg/kg SCP groups and extremly significantly decreased(p<0.01)in 100 mg/kg SCP group. The level of GSH was significantly increased(p<0.05)and the level of MDA was reduced(p<0.05)in liver tissue in 50 and 100 mg/kg SCP groups. SCP could improve hepatocyte degeneration and necrosis,inflammatory infiltration and inhibit hepatocyte apoptosis. Compared with the MOD group,SCP pretreatment could significantly decrease the expressions of p-JNK(p<0.05),decrease significantly Cleaved caspase-3 and Bax,and increase Bcl-2 protein expression(p<0.01). Conclusion:SCP has a protective effect on APAP-induced liver injury in mice,and its underlying mechanism might be related to the inhibition of hepatocyte apoptosis.
