高效液相色谱法测定食品中丙酸及其盐类

Determination of Propionic Acid and Its Salts in Food by High Performance Liquid Chromatography

  • 摘要: 本文通过不同沉淀剂、提取条件、提取方式对前处理方法进行了优化,同时考察了不同色谱柱和流动相条件对分离的影响,建立食品中丙酸及其盐类的高效液相色谱检测方法。试样经水提取,采用亚铁氰化钾-乙酸锌沉淀剂沉淀蛋白和杂质,以10 mmol/L磷酸氢二氨(pH3.0)的溶液为流动相,Atlantis T3 C18色谱柱进行分离,二极管阵列检测器检测,外标法定量。本方法丙酸在5.0~500.0 μg/mL之间线性关系良好(R2≥0.999),平均回收率在85.1%~109.4%,相对标准偏差均小于3.78%。方法检出限(S/N≥3)为0.02 g/kg,定量限(S/N≥10)为0.05 g/kg。该方法具有前处理简单、快速、准确等优点,适用于大批量样品中丙酸的测定,为企业和检验机构对食品中丙酸的监测提供了技术支持。

     

    Abstract: In this paper,the pretreatment methods were optimized by different precipitators,extraction conditions and extraction methods,and the effects of different chromatographic columns and mobile phase conditions on separation were investigated. A method for the analysis of propionic acid and its salts in food by high performance liquid chromatography(HPLC)was established. The samples were extracted with water and precipitated with ferrous potassium cyanide-zinc acetate precipitator to precipitate proteins and impurities. The propionic acid and its salts were separated on an Atlantis T3 C18 column using 10 mmol/L diammonium hydrogen phosphate solution as mobile phase,and detected with diode-array detector by external standard method. The method showed that the linearity of propionic acid was satisfied with the good correlation coefficients(R2≥0.999),in the concentration ranges of 5.0~500.0 μg/mL. The average recoveries for the target compound were between 85.1%~109.4%,with RSDs less than 3.78%. The limit of detection(S/N≥3)was 0.02 g/kg,and the limit of quantitative(S/N≥10)the was 0.05 g/kg. This method has the advantages of simple,rapid,and accurate pretreatment,is suitable for the determination of propionic acid and its salts in large quantity food samples. The developed method can provide strong technical support for enterprise quality control and government regulation.
