灵芝三萜酸改善小鼠睡眠作用研究

Hypnotic Effects of Ganoderma lucidum Triterpene Acids in Mice

  • 摘要: 目的:研究灵芝三萜酸对小鼠睡眠行为的影响。方法:将50只ICR雄性小鼠分为5组,分别为溶剂对照组、地西泮组、灵芝三萜酸高剂量组(T100 mg/kg)、灵芝三萜酸中剂量组(T50 mg/kg)、灵芝三萜酸低剂量组(T25 mg/kg)。各试验组每天经口给予5% 吐温80、2.5 mg/kg地西泮和不同剂量的灵芝三萜酸,30 d后运用直接睡眠实验、延长戊巴比妥钠睡眠时间实验、戊巴比妥钠阈下剂量催眠实验、巴比妥钠睡眠潜伏期实验评价灵芝三萜酸对小鼠睡眠的影响。结果:灵芝三萜酸灌胃给予30 d,对小鼠体重增长无影响;灵芝三萜酸低、中、高剂量组对小鼠均无直接促进睡眠的作用;在延长戊巴比妥钠睡眠时间实验中,与溶剂对照组相比,T25、T50、T100 mg/kg均能极显著延长戊巴比妥钠睡眠时间(p<0.01),T50、T100 mg/kg组睡眠潜伏期可显著缩短(p<0.05);在戊巴比妥钠阈下剂量催眠实验中,T100 mg/kg剂量组的入睡率强于地西泮2.5 mg/kg组,表明其对戊巴比妥钠阈下剂量小鼠的催眠有协同作用;在巴比妥钠睡眠潜伏期实验中,灵芝三萜酸各剂量组的睡眠潜伏期极显著缩短(p<0.01),100 mg/kg剂量的睡眠潜伏期极显著短于阳性对照组(p<0.01)。结论:灵芝三萜酸具有改善睡眠的功能。

     

    Abstract: Objective:To study effect of Ganoderma lucidum on the sleeping improvement in mice. Methods:50 ICR male mice were divided into 5 groups,which were solvent control group,diazepam group,and Ganoderma lucidum triterpene acids with high-dose group(100 mg/kg),medium-dose group(T50 mg/kg),and low-dose group(T25 mg/kg). Each test group was orally administered 5% tween 80,2.5 mg/kg diazepam and different doses of Ganoderma lucidum triterpenic acid. After 30 days,the effects of Ganoderma lucidum triterpenic acid on the direct sleep test,the extending test with sodium pentobarbital,the hypnosis test under subthreshold dose of sodium pentobarbital,and the incubation period test with barbital sodium were stuides. Results:The Ganoderma lucidum triterpenic acid was administered orally for 30 days,which had no effect on the weight gain of mice. The Ganoderma lucidum triterpene acids had no significantly direct sleep function on mice. The triterpene acids significantly not only prolonged pentobarbital-induced sleeping time at the doses of 25,50,and 100 mg/kg(p<0.01),but also reduced sleep latency at the doses of 50 and 100 mg/kg(p<0.05),compared to solvent control group. In the hypnosis test under subthreshold dose of sodium pentobarbital,triterpene acids at 100 mg/kg significantly stronger than that of diazepam group(2.5 mg/kg),which showed synergistic effect on subthreshold dose hypnosis with sodium pentobarbital. In the barbituric sodium sleep latency test,the sleep latency of Ganoderma lucidum triterpenic acid group was significantly shortened(p<0.01). The high dose at 100 mg/kg had a stronger effect than diazepam group(p<0.01). Conclusion:Ganoderma lucidum triterpenic acid had the function of improving sleep.
