鸡枞菌粗蛋白超声辅助提取工艺优化及其抗氧化活性

Optimization of Ultrasonic-assisted Extraction Process of Crude Protein from Termitomyces albuminosus and Its Antioxidant Activity

  • 摘要: 以鸡枞菌为原料,以蛋白提取率为指标,采用单因素实验和正交实验,优化了超声波辅助提取鸡枞菌粗蛋白的工艺条件,并探讨了鸡枞菌粗蛋白对ABTS+·、DPPH·、O2-·自由基的清除能力。结果表明:鸡枞菌粗蛋白提取的最佳条件为超声功率400 W、超声温度70 ℃、超声时间1.5 h、pH11、料液比1:25 g/mL,此条件下蛋白质含量为86.525%±1.315%,蛋白提取率为67.322%±1.028%。鸡枞菌粗蛋白对ABTS+·、DPPH·和O2-·三种自由基均有清除作用,其IC50分别为46.322、61.176和151.250 mg/L,但自由基的清除能力明显低于同浓度VC的清除能力。因此,超声波辅助适用于鸡枞菌粗蛋白的提取,所得粗蛋白具有抗氧化活性,该研究可为鸡枞菌高值化加工与利用提供理论依据。

     

    Abstract: Using Termitomyces albuminsous as experimental materials,the optimum conditions of ultrasonic-assisted extraction from Termitomyces albuminsous with the extraction rate of protein as index were studied through the method of single factor and orthogonal experiments. The crude protein’s antioxidant activities of ABTS+·,DPPH· and O2-· radical scavenging assays were also investigated in this paper.The results showed that the optimum extraction conditions were ultrasonic power 400 W,ultrasonic temperature 70 ℃,ultrasonic time 1.5 h,pH11 and solid-to-solvent ratio 1:25 g/mL,respectively. The extraction rate of crude protein could reach up to 67.322%±1.028% and the content of protein was 86.525%±1.315% under these conditions. Termitomyces albuminosus crude protein had a certain antioxidant activity. The scavenging ability of ABTS+·,DPPH· and O2-· free radical IC50(half maximal inhibitory concentration)value were separately 46.322,61.176 and 151.250 mg/L. The free radical scavenging capacity of Termitomyces albuminosus protein was lower than that of VC’s ability to remove them. Therefore,the ultrasonic-assisted technology was suitable for crude protein extraction from Termitomyces albuminosus and the protein had antioxidant activity. The study provided a theoretical basis for high value processing of Termitomyces albuminosus.
