NaCl和pH对普通变形杆菌和蜂房哈弗尼菌碳源代谢能力的影响

Effects of NaCl and pH on Carbon Source Metabolism Ability of Proteus vulgaris and Hafnia alvei

  • 摘要: 本文以轻腌大黄鱼优势菌(普通变形杆菌和蜂房哈弗尼菌)为研究对象,利用BIOLOG分析了两种菌在5 ℃条件下,不同NaCl浓度和pH水平胁迫对其碳源代谢的影响,通过丰富度指数、孔平均颜色变化率及利用面积,结合主成分分析法评价其碳源代谢能力。结果表明,普通变形杆菌和蜂房哈弗尼菌利用的碳源种类均以糖类为主,其他依次为羧酸类、氨基酸类和己糖磷酸类。NaCl浓度对两株菌的碳源利用能力影响随其浓度升高而增强,NaCl增加到5%时,几乎完全抑制了两株菌对碳源的利用。pH在5.0~7.0范围内,其对两株菌的碳源利用抑制效应随pH降低而增大,但总体抑制效应不显著。结论:NaCl浓度的增加能够逐渐抑制两株菌的碳源代谢,增加到3%时几乎完全被抑制,在pH5.0~7.0的范围内,对两株菌的碳源代谢影响不大。

     

    Abstract: Proteus vulgaris and Hafnia alvei were spoilage organisms isolated from lightly-salted large yellow croaker at 5 ℃. To analyze the effects of environmental factors on the carbon source metabolism of Proteus vulgaris and Hafnia alvei,BIOLOG was used to analyze the carbon source metabolism of two strains under different NaCl concentration and pH conditions at 5 ℃. The carbon source metabolism ability of Proteus vulgaris and Hafnia alvei was evaluated by the number of positive wells,the average well colour development,the utilization area and principal component analysis. The results showed that,the two strains digested similar carbon sources,such as sugars,carboxylic acids,amino acids and hexose phosphates. The effects of NaCl on the metabolism of the two strains increased with the increase of its concentration,and the metabolism was almost completely inhibited under 5% NaCl concentration. By comparison,the effects of pH on the carbon source metabolism of two strains were not obvious. The inhibition effects of pH(5.0~7.0)on the carbon source metabolism of the two strains increased with the decrease of pH values,but the overall inhibition effect was not significant. Conclusion:The increase of NaCl concentration could gradually inhibit the carbon metabolism of the two strains. When NaCl concentration increased to 3%,it almost completely inhibited the strains. In the range of pH5.0~7.0,the carbon source metabolism of the two strains had little effect.
