大叶苦丁茶多酚对四氯化碳致小鼠肝损伤的预防作用

Preventive Effect of Large-leaved Kuding Tea Polyphenols on CCl4-Induced Hepatic Damage in Mice

  • 摘要: 为检测大叶苦丁茶多酚提取物(polyphenol of large-leaved Kuding tea,PLK)对CCl4诱导小鼠肝损伤的预防作用,将昆明雄性小鼠连续14 d给予低(50 mg/kg)、高(100 mg/kg)两个剂量的大叶苦丁茶多酚提取物及药物水飞蓟素(100 mg/kg)灌胃,然后腹腔注射0.8% CCl4建立动物急性肝损伤模型。从小鼠眼静脉丛取血液,将其处死后测定小鼠肝重、肝脏指数、血清指标和肝组织中氧化和炎症相关mRNA表达。实验结果表明PLK可以使肝损伤小鼠的肝指数降低;PLK能减少肝损伤小鼠血清中的AST(天门冬氨酸氨基转移酶)、ALT(丙氨酸氨基转移酶)、TG(甘油三酯)、TC(总胆固醇)的含量。病理学形态观察显示PLK能减少CCl4引起的肝组织中心静脉分化和肝细胞损伤。qPCR实验结果表明,PLK能够显著上调小鼠肝组织中Cu/Zn-SOD(铜锌超氧化物歧化酶)、Mn-SOD(锰超氧化物歧化酶)、IκB-α(核因子κB 抑制蛋白α)、CAT等抗氧化酶的 mRNA表达和下调COX-2(环氧化酶-2)、iNOS(诱生型一氧化氮合酶)、TNF-α(肿瘤坏死因子-α)、NF-κB(核因子κB)、IL-1β(白细胞介素-1β)的表达(p<0.05)。PLK对CCl4引发的肝损伤有较好的预防作用,且与药物水飞蓟素有相似效果,具有良好的功能性作用。

     

    Abstract: The preventive effects of polyphenols of large-leaved Kuding tea(PLK)on CCl4-induced hepatic damage in mice were determined in this study. The Kunming male mice were given low(50 mg/kg)and high(100 mg/kg)concentration of PLK,and drug of silymarin(100 mg/kg)by gavage for 14 d,then the animal model of acute liver injury was established by intraperitoneal injection of 0.8% CCl4.The blood was collected from the ophthalmic venous plexus of mice. Liver weight,liver index,serum markers and expression of oxidative and inflammatory related mRNA genes in liver tissues were measured after the mice were executed. The experiment results showed that PLK could reduce the liver index in hepatic damage mice,meanwhile,and PLK also could reduce the serum aspartate aminotransferase(AST),alanine aminotransferase(ALT),triglyceride(TG),total cholesterol(TC)levels. Pathological observation also showed that PLK could effectively reduce the hepatictissue destruction and hepatocellular necrosis caused by CCl4.The qPCR experiment results showed that PLK could significantly increase copper/zinc-superoxide dismutase(Cu/Zn-SOD),manganese-superoxide dismutase(Mn-SOD),nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor-alpha(IκB-α),catalase(CAT)mRNA expressions and decrease cyclooxygenase-2(COX-2),inducible nitric oxide synthase(iNOS),tumor necrosis factor-alpha(TNF-α),nuclear factor-kappa B(NF-κB),interleukin 1 beta(IL-1β)expressions of hepatic tissues in mice compared with the mice in control group(p<0.05). PLK had good preventive effect on CCl4 induced hepatic damage,and PLK had the similar effects with silymarin,which has good functional effect.
