美洲大蠊脱脂膏中总糖和还原糖含量的测定

Determination of Total Sugar and Reducing Sugar in Degreasing Cream from Periplaneta americana

  • 摘要: 目的:采用不同方法测定美洲大蠊脱脂膏中总糖含量,优选出测定美洲大蠊脱脂膏中总糖含量的方法,并建立其还原糖含量测定方法。方法:以葡萄糖为标准品,分别采用苯酚-硫酸法和硫酸-蒽酮法测定总糖含量,采用二硝基水杨酸法测定还原糖含量。结果:苯酚-硫酸法标准品浓度在0.004~0.100 mg/mL范围内呈良好的线性关系,平均加样回收率为98.7%,RSD为2.0%,R2=0.9996。硫酸-蒽酮法标准品浓度在0.02~0.20 mg/mL范围内有良好的线性关系,平均加样回收率为97.7%,RSD为2.6%,R2=0.9997。二硝基水杨酸法标准品浓度在0.004~0.040 mg/mL范围内呈良好的线性关系,平均加样回收率为96.8%,RSD为1.6%,R2=0.9996。结论:苯酚-硫酸法可用于脱脂膏中总糖含量的测定,二硝基水杨酸法可用于脱脂膏中还原糖含量的测定。

     

    Abstract: Objective:The content of polysaccharides in the degreasing cream of Periplaneta americana was determined by different methods in order to select an accurate and convenient method for determination of total sugar content in Periplaneta americana extract and establish a method for determining reducing sugar content. Methods:Phenol-sulfuric acid method and anthrone colorimetry were used to determine the total sugar content. DNS method was used to determine the reducing sugar content. Results:The glucose was in a good linear relationship in the range of 0.004~0.100 mg/mL (R2=0.9996), and the recovery rate was 98.7% (RSD=2.0%) by phenol-sulfuric acid method. The glucose was in a good linear relationship in the range of 0.02~0.20 mg/mL (R2=0.9997), and the recovery rate was 97.7% (RSD=2.6%) by anthrone colormetry. The glucosewas in a good linear relationship in the range of 0.004~0.040 mg/mL (R2=0.9996), and the recovery rate was 96.8% (RSD=1.6%) by DNS method. Conclusion:The phenol-sulfuric acid method could be used to determine the total sugar content. The DNS method could be used to determine the reducing sugar content.
