酶解鸢乌贼组织蛋白制备酸溶性肽的工艺优化

Optimization on the Preparation Process of Acid Soluble Peptides from Symplectoteuthis oualaniensis Tissue Protein by Enzymolysis

  • 摘要: 为有效利用鸢乌贼资源,对酶解鸢乌贼组织蛋白制备酸溶性肽的工艺进行了研究。比较了不同蛋白酶对鸢乌贼组织蛋白水解能力的差异,确定胰蛋白酶为水解鸢乌贼组织蛋白的最佳蛋白酶类型。先后采用单因素和响应面试验对酶解工艺条件进行了系统的研究及优化。确定了胰蛋白酶水解鸢乌贼组织蛋白的最佳工艺条件为:蛋白浓度3%,加酶量4210 U/g蛋白,pH9.23,酶解温度52.3 ℃,酶解时间5 h。在优化的工艺条件下,酸溶性肽的得率可以达到(74.02%±0.41%)。此外,氨基酸分析结果表明,经酶解得到的酸溶性肽基本保持了原鸢乌贼组织蛋白的氨基酸组成,8种必需氨基酸含量丰富,约占氨基酸总量的38%。试验结果表明,采用胰蛋白酶水解鸢乌贼制备酸溶性肽的工艺具有可行性。

     

    Abstract: To improve the utilization and economic value of Symplectoteuthis oualaniensis resource, the enzymatic hydrolysis process of Symplectoteuthis oualaniensis tissue protein for the production of acid soluble peptides was investigated. First of all, the different types of proteases on hydrolysis of Symlectoteuthis oualaniensis tissue protein were compared, and trypsin was determined as the preferable protease suitable for the hydrolysis process. Then, the hydrolysis process conditions were analyzed and optimized by single factor experiments and response surface methodology. Finally, the optimum process conditions were determined:protein concentration 3%, enzyme dosage 4210 U/g, pH9.23, temperature 52.3℃, reaction time 5 h. Under the optimized conditions, the yield of acid soluble peptide could reach (74.02%±0.41%). In addition, the results of amino acid composition showed that the acid soluble peptides ccould effectively maintain the amino acid composition of Symlectoteuthis oualaniensis tissue protein, 8 essential amino acids were abundant, accounting for about 38% of total amino acids. All of these results showed that the preparation process of acid soluble peptides from hydrolysis of Symplectoteuthis oualaniensis tissue protein by protease was feasible.
