厚皮甜瓜果实对链格孢侵染抵抗能力的差异

Resistance difference of muskmelon fruit on infection of Alternaria alternata

  • 摘要: 以伽师瓜和86-1甜瓜为实验材料,研究贮藏期间甜瓜对病原菌抵抗能力的差异。用打孔器在果实赤道部位等距离打孔6个,每孔接种20 μL链格孢,于7 ℃冷库贮藏,研究果皮与果肉病斑直径、几丁质酶、β-1,3-葡聚糖酶、苯丙氨酸解氨酶、肉桂酸羟化酶和4-香豆酸-辅酶A连接酶活性的变化。结果表明:贮藏期间,86-1甜瓜果实病斑直径大于伽师瓜,且果肉病斑直径大于果皮;甜瓜接种链格孢后,果实病程相关蛋白及苯丙烷代谢酶活性均高于对照,在贮藏期间整体呈现先升高后下降的变化趋势。研究表明,伽师瓜果实抗病相关酶活性高于86-1甜瓜,果皮酶活性高于果肉,伽师瓜的抗病能力强于86-1甜瓜,果皮的抗病能力强于果肉。

     

    Abstract: Take Jiashi muskmelon and 86-1 muskmelon as the experimental materials,the difference of resistance ability of muskmelon to pathogens during storage was studied in this paper. Six holes were made with a hole punch at regular distance on the equatorial part of fruit. And every hole inoculated with 20 μL A.alternata,then the muskmelon was stored in cold storage at 7 ℃. The lesion diameter of pericarp and sarcocarp were measured. The activities of chitinase,β-1,3-glucanase,phenylalanine ammonia-lyase,cinnamic acid hydroxylase and 4-coumaric acid-coenzyme A ligase were studied. The results indicated that during the storage period,the lesion diameter of 86-1 muskmelon was greater than that of Jiashi muskmelon,and the sarcocarp lesion diameter was greater than the pericarp. After inoculated with the A.alternata,both the pathogenesis-related proteins and the phenylpropanoid metabolizing enzymes were higher than CK,which showed the trend of rising first and then decreasing during the storage period. According to the research,the activities of disease-related enzymes in Jiashi muskmelon were higher than that of 86-1 muskmelon,which resulted the former was more resistant to disease than the latter. Pericarp enzyme activities were higher than the sarcocarp,so the pericarp resistance was stronger than the sarcocarp.
