高效液相色谱法检测乳中β-内酰胺酶抑制剂他唑巴坦

Determination of β-lactamase inhibitor Tazobactam in milk by high performance liquid chromatography

  • 摘要: 建立了乳中β-内酰胺酶抑制剂他唑巴坦的高效液相色谱检测方法。样品采用乙腈作为沉淀蛋白溶剂,正己烷作为脱脂溶剂。最佳色谱条件:色谱柱为Hypersil ODS-2 C18(4.6 mm×250 mm,5 μm),流动相为A:B为(10:90),检测波长为213 nm。结果表明,他唑巴坦在5~200 μg/mL的浓度范围内具有良好的线性关系,且相关系数是0.9991,采用空白基质匹配标准溶液的方法进行标准校正,显示他唑巴坦的平均回收率在88.40%~94.17%之间。该方法具有操作简单,灵敏度较高,结果准确等特点,适用于乳中他唑巴坦的检测。

     

    Abstract: An analytical method was developed and verified for the determination of β-lactamase inhibitor Tazobactam in milk by high performance liquid chromatography.The sample was deproteinized by acetonitrile and degreased by n-hexane. The chromatographic conditions was chosen Hypersil ODS-2 C18 chromatographic column using A∶B(10∶90)as mobile phases with 213 nm detection wavelength. The results of the calibration curve showed perfect linear in the range of 5~200 μg/mL and the correlation coefficient was 0.9991.Matrix-matched calibration standard was used for the quantification and showed the average recoveries ranges from 88.40%~94.18%. With the advantages of simple operation,high sensitivity and accurate results,the method was suitable for the detection of Tazobactam in milk.
