甜柿果实多酚类物质提取工艺的优化及其抗肺癌细胞生长的活性研究

Extraction conditions optimization and inhibition the growth of human lung cancer in vitro of polyphenols from the fruit of Diospyros kaki Thunb.

  • 摘要: 为确定罗田甜柿果实中多酚的最佳提取工艺条件及其抗肿瘤活性,在单因素实验的基础上,通过二元多次回归拟合对提取时间、提取温度和乙醇体积分数进行响应面优化后,得到甜柿果实多酚的最佳提取工艺条件:以体积分数为75%的乙醇在70℃的温度条件下提取2.9 h,柿果多酚的提取率为3.184 mg/100 g鲜果重。通过MTT实验,发现柿果多酚对CL1-5细胞的半抑制浓度(IC50)值为73.8 μg/mL,证实甜柿果实多酚对人肺癌细胞系CL1-5具有抑制作用。

     

    Abstract: The optimal extraction conditions and anti-tumor activities of polyphenols from the fruit of Diospyros kaki Thunb. were studied in this article. The optimal polyphenols extraction conditions were acquired based on the single factor experiment,binary multiple regression fitting and Box-Behnken multimodel experiment,and optimized by response surface methodology. The optimum conditions for polyphenols extraction from persimmon were as follows:extraction time 2.9 h,temperature 70℃,ethanol concentration 75%. Under the optimum conditions,the extraction ratio of polyphenols was 3.184 mg/100 g. Based on the result of MTT,it is found that persimmon polyphenols has an inhibitory effect on CL1-5 cells with the IC50 value of 73.8 μg/mL,and the result showed polyphenols have an inhibitory effect on human lung cancer cell line CL1-5.
