pH控制策略及pH反馈流加培养法提高罗氏产碱杆菌的聚羟基丁酸产量

pH control strategy and pH feedback flow culture method used to improve the production of polyhydroxybutyrate produced by Roche alcaligenes sp

  • 摘要: 通过控制不同的pH以及利用pH反馈来控制培养基的流加方式等方法提高罗氏产碱杆菌的生物量及聚羟基丁酸(PHB)的产量。根据罗氏产碱杆菌在分批培养及不同流加培养基的实验中,确定了pH反馈流加培养基的方法。利用该方法在发酵过程中补加400 g/L的葡萄糖和100 g/L的酵母浸粉,并且将该过程的pH控制在6.7~6.9。在此条件下罗氏产碱杆菌的干重达到22.52 g/L,PHB浓度可到13.61 g/L。罗氏产碱杆菌的生物量及PHB浓度分别提高了12.44%和5.07%,结果表明采用pH反馈控制培养基的流加方式可以提高PHB的产量。

     

    Abstract: In this passage,the biomass and production of polyhydroxybutyrate(PHB)were studied by controlling different pH values and using pH feedback to control the flow adding mode of medium. According to the experiments of batch culture and different culture medium,the pH feedback medium was confirmed. With this method,400 g/L glucose and 100 g/L yeast dip powder were added to the fermentation process,and the pH of the process was controlled at 6.7~6.9. Under these conditions,the dry weight of Roche alcaligenes sp was 22.52 g/L,and the yield of PHB reached 13.61 g/L. The biomass and PHB yield of Bacillus alcaligenes increased by 12.44% and 5.07%. The results show that PHB can be improved by the pH feedback control medium.
