石菖蒲中高纯度β-细辛醚和α-细辛醚的制备

Preparation of high purity β-asarone and α-asarone from Acorus tatarinowii Schott

  • 摘要: 建立了采用高压半制备液相色谱制备石菖蒲中β-细辛醚和α-细辛醚高纯单体化合物的方法。首先以石菖蒲为原料制备石菖蒲挥发油,然后采用高压半制备液相色谱,设定流动相为甲醇∶水=65∶35(v/v),对β-细辛醚和α-细辛醚进行精制纯化。产物的结构采用EI-MS、1H-NMR和13C-NMR进行鉴定,而且它们的纯度分别达到99.43%和99.08%。该方法操作简单、制备量大、产物纯度高,可用于石菖蒲中高纯度β-细辛醚和α-细辛醚对照品的制备。 

     

    Abstract: To establish a method for preparation of high purity β-asarone and α-asarone from Acorus tatarinowii Schott by high pressure semi-preparative liquid chromatography, its volatile oil was prepared from Acorus tatarinowii Schott, and a semi-preparative high performance liquid chromatography was used to refine and purify β-asarone and α-asarone with a solvent composed of methanol and water ( 65∶ 35, v/v) .The structures of product were identified by EI-MS, 1H-NMR and13C-NMR, and their purities reached 99.43% and 99.08%, respectively. The method is easily operated, with high purity and preparation amount.It is suitable for high purity β-asarone and α-asarone reference material preparation from Acorus tatarinowii Schott.
