双水相体系萃取蓝莓多糖的分配平衡研究

Optimization of two-phase extraction of polysaccharide from blueberry

  • 摘要: 采用超声水提法提取蓝莓多糖,再用双水相法进行纯化。以药液百分数、K2HPO4浓度、PEG6000浓度为单因素,以下相多糖的萃取率为指标,应用响应面法优化蓝莓多糖的双水相萃取工艺。结果表明,当药液百分数60%,PEG6000 16%,K2HPO4 16.9%,下相蓝莓多糖得率最高,可达95.03%;且多糖纯度明显高于传统的水提醇沉法。所得到模型高度显著(R2=0.9920),拟合性好,可用于蓝莓多糖的萃取。 

     

    Abstract: The polysaccharide was extracted by ultrasonic water extraction,then purified by aqueous two phase. With the percentage of solution,the concentration of K_2HPO_4 and the concentration of PEG6000 as the single factor,and extraction rate of polysaccharides from the lower phase as the index,the double phase extraction technology of blueberry polysaccharide was optimized by response surface methodology. The results showed that the percentage of liquid medicine 60%,PEG6000 16%,K_2HPO_4 16.9%,the next phase of blueberry polysaccharide yield was the highest,up to 95.03%.And the purity of polysaccharide was significantly higher than that of the traditional water extraction and alcohol precipitation method.The model was highly significant( R~2= 0.9920),good fitting,can be used for extraction of polysaccharides from blueberry.
