南极磷虾抗冻蛋白的特异性亲和吸附提取研究

Extraction of the antifreeze proteins from Antarctic Krill( Euphausia superba) by specific affinity adsorption

  • 摘要: 本文以南极磷虾蛋白为原料,以南极磷虾抗冻蛋白(Antifreeze proteins,AFPs)的得率为指标,开展了特异性亲和吸附法提取南极磷虾AFPs的工艺研究。本文在单因素实验的基础上,通过响应面优化实验研究样品浓度、吸附温度、吸附时间、吸附次数对南极磷虾AFPs得率和热滞活性(Thermal Hytersis Activity,THA)的影响。结果表明,南极磷虾AFPs的最优提取条件为:样品浓度1.0 mg/m L、吸附温度-1.70℃、吸附时间为11 h、吸附次数为2次。在上述条件下预测南极磷虾AFPs的得率为29.34%,THA为0.0805℃;验证值与预测值之间的误差分别为2.66%和1.86%。这表明该模型可以用于预测南极磷虾AFPs的提取优化。本文的研究结果为南极磷虾AFPs的深入研究提供技术支撑。 

     

    Abstract: In this paper,the key factors and optimal conditions of extraction of Antarctic Krill AFPs( Antifreeze proteins,AFPs) were researched by specific affinity adsorption. On the basis of single factor experiment,sample concentration,adsorption temperature,adsorption time and adsorption number were optimized by response surface methodology with the AFPs extraction yield and THA as response value. The results indicated that the optimal extraction process was sample concentration of 1.0 mg / m L,adsorption temperature of-1.70 ℃,adsorption time of 11 h,the adsorption number of 2. Under the optimal conditions,the model optimum solution would be29.34% and 0.0805 ℃. The relative errors were 2.66% and 1.86% between actual and model value. The fitted mathematical models for the extraction yield of Antarctic Krill AFPs and its THA had a good feasibility.
