板栗壳生药研究及有效成分没食子酸、绿原酸、芦丁含量测定

Pharmacognostical studies of Chestnut Epicarp and determination of active ingredients including gallic acid,chlorogenic acid and rutin

  • 摘要: 采用性状鉴别、组织切片鉴别、粉末鉴别对板栗壳进行生药研究。建立HPLC法同时测定板栗壳中没食子酸、绿原酸和芦丁含量,色谱条件为C18柱,甲醇-0.4%磷酸为流动相梯度洗脱,检测波长275 nm,流速1 m L/min,柱温30℃。结果表明,板栗壳横切面、纵切面果皮细胞特点、中果皮的石细胞群和石细胞环带、内果皮较多的非腺毛、种脊部位的三角形维管束及粉末中果皮细胞、石细胞、非腺毛特征均可作为板栗壳的生药鉴别特征,为板栗壳生药学质量标准研究奠定基础。测定11批云南不同产地板栗壳样品中没食子酸含量为0.3280.362 mg/g,绿原酸含量为0.1290.141 mg/g,芦丁含量为0.1800.230 mg/g。该含量测定方法操作简单,结果准确,专属性强,为板栗壳质量标准的建立提供依据。 

     

    Abstract: Pharmacognostical studies of Chestnut Epicarp were carried out by using morphological identification,tissue slice identification and powder identification. Gallic acid,chlorogenic acid and rutin were simultaneous determined by HPLC equipped with Hypersil ODS2 column and UV detection was used,the detection wavelength was set at 275 nm.The mobile phase was consisited of methanol and 0.4% phosphoric acid with gradient elution.The flow rate was 1.0 m L / min and the column temperature was at 30 ℃.The results showed that these features in transverse and longitudinal sections of Chestnut Epicarp,pericarp cell,stone cells groups and stone cell ring of sarcocarp,a large number of non-glandular hairs of endocarp and triangular vascular bundle located at the raphe site could be used as Chestnut Epicarp pharmacognostical characteristics,a dditionally,the powder characteristics of pericarp cell,stone cells and non-glandular hairs were pharmacognostical characteristics of Chestnut Epicarp,which laid the foundation to further study pharmacognostical quality standard of chestnut epicarp. The content range of gallic acid was 0.328 ~ 0.362 mg / g,chlorogenic acid was 0.129 ~ 0.141 mg / g,and rutin was 0.180 ~0.230 mg / g in the chestnut epicarp produced at 11 different places in Yunnan. The method was simple,accurate and specific.It can provide basis for the establishment of quality standard of chestnut epicarp.
