激素诱导和非生物胁迫对虎杖根中白藜芦醇含量影响及虎杖根中白藜芦醇闪提工艺优化

Effect of phytohormones induced and abiotic stresses on resveratrol content and optimization of resveratrol flash-extraction in polygonum cuspidatum roots

  • 摘要: 用紫外辐射和水杨酸、乙烯利、H2O2溶液对新鲜虎杖根进行处理,探究其对新鲜虎杖根中白藜芦醇含量的影响。采用闪式提取法从虎杖根中提取白藜芦醇,在单因素实验的基础上进行正交实验探究最佳提取工艺条件,用HPLC对白藜芦醇进行定性定量分析。结果表明:紫外辐射15 min,暗处理24 h后虎杖根中白藜芦醇的含量达到最大值,较辐射前提高了72%。10 mmol/L水杨酸溶液浸泡24 h后虎杖根中的白藜芦醇含量提升到原来的4倍多。5%H2O2溶液浸泡24 h后虎杖根中白藜芦醇含量上升了34%。而乙烯利溶液浸泡则会降低白藜芦醇的含量。闪式提取法提取虎杖根中白藜芦醇的最佳工艺条件是:提取温度50℃,提取时间60 s,乙醇浓度80%,料液比例1∶30 g/m L,在此条件下,白藜芦醇得率为0.1877%。 

     

    Abstract: The effects of UV lights,salicylic acid,ethephon and H2O2 on resveratrol induction in fresh polygonum cuspidatum roots were studied. The effects of extraction temperature,extraction time,ethanol concentration and solid-liquid ratio on resveratrol content were investigated.Based on the single factor experiments,the optimization of resveratrol flash-extraction process was carried by the orthogonal test.And the resveratrol content was analyzed by HPLC. The results showed that the resveratrol content in polygonum cuspidatum could reach its maximum( improve 72%) after UV radiation 15 min and dark treatment 24 h.Its content increased by more than four times after soaking in the 10 mmol / L salicylic acid for 24 h and it increased by 34% after soaking in 5% H2O2 for 24 h.But ethephon did not work. The best flash-extraction conditions were as follows: extraction temperature 50 ℃,extraction time 60 s,ethanol concentration 80% and solid-liquid ratio 1 ∶ 30 g / m L.Under these conditions,the extraction rate of resveratrol was 0.1877%.
