凝结芽孢杆菌Liu-g1产中性蛋白酶的发酵培养基及条件优化

Optimization of culture medium and fermentation conditions for neutral protease produced by Bacillus coagulans Liu-g

  • 摘要: 为提高一株产中性蛋白酶凝结芽孢杆菌(Bacillus coagulans)Liu-g1发酵液中蛋白酶活力,采用单因素和正交实验研究不同培养基配方和不同发酵条件对凝结芽孢杆菌Liu-g1产中性蛋白酶活力的影响。结果表明:发酵培养基最优配方为大豆粕1%、玉米淀粉4%、碳酸钙0.6%;在初始p H7.5,装液量30 m L/250 m L,发酵温度45℃,发酵时间72 h优化条件下发酵液中蛋白酶活力最高,可达243.874 U/m L,为优化前的5.8倍。 

     

    Abstract: In order to improve the activity of neutral protease produced by Bacillus coagulans Liu-g1,single factor test and orthogonal test on Bacillus coagulans Liu-g1 protease activity were carried out. The results showed that the optimal culture medium formula was 1% of soybean powder,4% of corn starch and 0.6% of calcium carbonate.The results also indicated that the best initial p H was 7.5,fermentation volume of 30 m L /250 m L,incubation temperature at 45 ℃,fermentation time for 72 h. The protease activity reached 243.874 U / m L in such conditions,5.8 times the amount of initial conditions.
