利用酶标仪快速批量测定橡胶草中菊糖的含量

Rapid batch determination of the inulin in Taraxacum kok- saghyz Rodin by enzyme mark instrument

  • 摘要: 以橡胶草根为研究对象,采用3,5-二硝基水杨酸法(DNS法),利用酶标仪为检测仪器,建立了橡胶草中菊糖的快速测定方法。结果表明,酶标仪检测橡胶草根中总糖和还原糖含量的最佳实验条件:波长为550 nm、DNS显色剂用量为0.8 m L、沸水浴显色时间为8 min。同时,获得此条件下回归方程为A=0.6872C-0.0215(R2=0.9982),线性范围为02.2 mg/m L,加样回收率为96.67%102.33%。该方法灵敏、稳定、重现性好,可在短时间内完成多个样品同时测定,该方法所得结果略高于分光光度计法。 

     

    Abstract: In order to establish the rapid batch determination method,which could be used in the determination of the inulin in Taraxacum kok- saghyz Rodin root by enzyme mark instrument,3,5- dinitrosalicylic Acid Colorimetry( DNS) was used as the mainly methods. The results showed that the optimum conditions as follows,the wave length was 550 nm,the volume of color solution was 0.8 m L,the coloration time was 8 min.Under this condition,the calibration curve of this method had a liner range from 0 to 2.2 mg / m L with a regression equation of A = 0.6872C-0.0215( R~2= 0.9982),and the spotting recovery rate was from 96.67% to 102.33%.The method was sensitive,stable and reproducibility to operate,and it was suitable to be used for rapidly and quantitatively analysis of inulin in Taraxacum kok- saghyz Rodin.The results of the inulin in Taraxacum kok- saghyz Rodin,which had been detected by enzyme is more higher than by spectrophotometry.
