八角茴香总黄酮抗氧化活性研究

Study on antioxidative effect of total flavonoids from Illicium verum

  • 摘要: 本实验采用Fenton法测定了八角茴香总黄酮(FIVR)对羟基自由基(·OH)和超氧阴离子自由基(O2-·)的清除作用,并与维生素C(VC)进行了比较,采用FRAP法测定了总抗氧化能力(T-AOC);采用腹腔注射D-半乳糖建立亚急性衰老小鼠模型。动物随机分为正常对照组、模型对照组、阳性对照组、FIVR高、中、低剂量组(200、100、50 mg/g·d)。实验48 d后处死动物,称量体重,计算器官指数,测定小鼠血清、肝脏和脑组织中的超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活力。结果发现,八角茴香总黄酮(FIVR)对·OH、O2-·的清除率和T-AOC均随着浓度的增加而上升;模型组小鼠体重、肝脏、脑指数及血清、肝、脑组织中SOD和GSH-Px活性均明显低于正常组,建模成功;而FIVR各给药组及VC组小鼠体重、肝脏、脑指数及血清、肝、脑组织中SOD和GSH-Px活力均高于模型组。说明八角茴香总黄酮(FIVR)具有明显的抗氧化活性。 

     

    Abstract: The clearance effect of total flavonoids from Illicium verum( FIVR) on ·OH,O2-· were determined using Fenton method and compared with that of vitamine C.Total antioxidant capacity( T- AOC) were determined using FRAP method to evaluate FIVR in vitro antioxidant activity. The aging mice model was established by the intraperitoneal injection of D- galactose.The mice were randomly divided into normal control group,model group,positive control group,FIVR high,medium and low- dose groups( 200,100,50 mg / g·d). After administrations for48 d,the body weight of mice were determined,then the organ coefficients were counted. The content of SOD,GSH- Px of liver,brain tissue and in the blood was detected. From FIVR antioxidant experiment in vitro,the clearance rate of ·OH,O2-· and T- AOC were increased with the increase of concentration.The body weight of mice,liver index,brain index and the activity of SOD,GSH- Px in liver tissue,brain tissue of model group were lower than normal group,which proved successful model; each FIVR dose group mice weight,liver index,brain index and the activity of SOD,GSH- Px in liver tissue and brain tissue were higher than model group. This study shows that FIVR has obvious antioxidation activity.
