响应曲面法优化蜗牛酶辅助提取黑木耳多糖工艺

Optimization of the snailase assistant extraction process of polysaccharides from Auricularia Auricula by response surface methodology

  • 摘要: 以黑木耳超微粉为原料,采用蜗牛酶酶解法,在单因素实验的基础上,采用响应曲面法对黑木耳多糖的酶提工艺进行优化。结果表明:酶解时间2.5 h,酶解温度为55.0℃,p H为6.8,酶的添加量为1.3%。在此条件下,多糖得率为31.1%,与理论预测值30.34%的相对误差约为0.76%,重现性良好。说明通过响应曲面优化得到的回归方程具有一定的实践指导意义。 

     

    Abstract: In this paper,superfine powder of Auricularia Auricular was selected as raw material,and polysaccharide from Auricularia Auricular was extracted by enzyme hydrolysis by snailase. On the basis of the single factor experiment,extraction condition was optimized through method of response surface. The results showed that:the optimal extraction condition was enzymatic hydrolysis time 2.5 h,enzymatic hydrolysis temperature55.0 ℃,p H6.8 and enzyme concentration 1.3%. Under above conditions,yield of polysaccharide reached31.1%,the relative average error between predicted yield and actual value was 0.76%,and the reproducible was reliable. So,it was proved that the response surface equation of optimized extracted condition derived some practical significance.
