曲拉复合乳酸菌增殖培养基的优化

Optimization of enrichment medium of Qula compound lactic acid bacteria

  • 摘要: 为了提高曲拉专用复合乳酸菌发酵液中的活菌数,通过基础培养基的筛选实验,氮源、碳源和生长因子的单因素实验,响应面实验设计对其增殖培养基成分进行优化。3株曲拉专用乳酸菌保加利亚乳杆菌(MGD1-3)、嗜热链球菌(MGB39-5)、植物乳杆菌(BM5152)间无拮抗作用,可以进行混菌培养,复合乳酸菌最佳基础培养基为MRS培养基。通过Box-Behnken实验及响应面分析确定碳源、氮源、吐温80和VB6的最佳添加量分别为:18.11、26.72、1.88和1.41 g/L,发酵菌株在优化后的MRS培养基中OD600值为2.173,菌体浓度达到了4.01×1010CFU/m L,活菌数增加了8.29倍。 

     

    Abstract: In order to improve viable Qula special compound lactic acid bacteria in the fermentation liquid,the single factor experiment of nitrogen sources,carbon sources and growth factors,response surface methodology were used to optimize the composition of enrichment medium. There were no antagonistic effects among MGD1- 3,MGB39- 5 and BM5152,so they can mix cultures. And it indicated that MRS medium was the most suitable for compound lactic acid bacterias growth.Box- Behnken test and response surface were analyzed to determine the optimum adding amount,and its optimal composition was carbon source 18.11 g / L,nitrogen source 26.72 g / L,Tween-80 1.88 g / L and VB61.41 g / L.The OD600 of strains cultured in the optimized MRS medium reached 2.173,the cell concentration in the optimized MRS medium reached 4.01 × 1010 CFU / m L,which was 8.29 times higher than observed in the initial medium.
