响应面法优化萌发菌HL-003胞内多糖提取工艺

Optimization of extraction conditions of intracellular polysaccharides from HL-003 using response surface methodology

  • 摘要: 为确定萌发菌HL-003胞内多糖提取工艺的最佳条件,以萌发菌HL-003胞内多糖提取得率为考察指标,在单因素实验的基础上,采用响应面法对主要提取工艺参数进行优化,并得到回归模型。结果表明:萌发菌HL-003胞内多糖提取工艺的最优条件参数为超声波处理时间19 min、液料比42 m L/g、浸提温度72℃、提取时间2.2 h,最大提取得率达2.03%。实验证明模型拟合程度良好,误差较小。 

     

    Abstract: Response surface methodology was used to optimize main process parameters for the extraction of polysaccharides from HL-003's intracellular broth based on polysaccharide extraction rate of one- factor tests.A regression model equation was fitted.The optimum extraction conditions were found to be ultrasound time 19 min,liquid- solid ratio 42 m L / g,extraction temperature 72 ℃ and extraction time 2.2 h.The extraction rate of polysaccharides under these conditions was up to 2.03%.The fitted regression model had excellent goodness of fit and showed lower prediction error.
