海带中岩藻多糖的抗衰老活性及构效关系研究

Study on the effect of anti- aging and structure- function relationship of fucoidan from kelp

  • 摘要: 研究海带中岩藻多糖的抗衰老活性及构效关系。将海带中岩藻多糖(FSP)经分离纯化得到三个FSP分离组分(FSP-1、FSP-2、FSP-3),对3个组分进行了元素分析并分别测定各组分分子量及其单糖组成;通过对D-半乳糖致衰老的小鼠给药,并测定一些与衰老相关的生化指标,以研究海带中3个分离组分的抗衰老活性。结果表明FSP-1、FSP-2、FSP-3治疗组与模型组比较,FSP-2与FSP-3均使衰老小鼠胸腺指数与脾脏指数明显回升(p<0.01),且FSP-3作用效果更为明显。3个组分均能显著升高小鼠血清中超氧化物歧化酶活性,降低丙二醛含量,以及升高全血中谷胱甘肽过氧化物酶活性(p<0.05或p<0.01)。说明FSP中3个组分均具有抗衰老活性,各分离组分的抗衰老活性强弱与其硫酸基含量、分子量大小和单糖构成有关。 

     

    Abstract: The effect of anti- aging and structure- function relationship of fucoidan from kelp was studied. The fucoidan was extracted from kelp and FSP- 1,FSP- 2,FSP- 3 were purified. Elemental analysis,gel permeation chromatography coupled with multi- angle laser light scattering( GPC- MALLS) and GC analysis were applied to investigate the three components. In order to study the effect of anti- aging of three components from kelp,administered medicine was given to rats with subacute senile model by D- galactose,then some indexes were determined. Compared treatment group with senile model group,indexes of thymus and spleen( p < 0.01) were promoted by FSP-2 and FSP- 3,moreover,the effect of FSP- 3 was more significant than FSP- 2. The activity of superoxide dismutase( SOD) and GSH- PX in blood were significantly promoted,but the content of malondialdehyde( MDA) was decreased by three components from kelp. Three components from kelp had the activity of anti- aging,and the activity of anti- aging was in relation with the content of sulfate groups,molecular mass and sugar composition of them.
