快速溶剂萃取联合高效液相色谱法测定不同产地酸枣仁中皂苷A、B的含量

Determination of jujuboside A and B in Zizyphi Spinosi Semen harvested from different places by using HPLC-ELSD combined with ASE

  • 摘要: 目的:建立酸枣仁皂苷A、B含量测定的前处理及分析方法,测定不同产地所产酸枣仁中皂苷A、B的含量。方法:采用快速溶剂萃取法(Accelerated Solvent Extraction,ASE)从酸枣仁中提取皂苷A、B,优化快速溶剂萃取法的提取条件以确定最佳条件,对蒸发光散射检测器(Evaporative Light Scattering Detector,ELSD)检测参数优化后,采用高效液相色谱(High Performance Liquid Chromatography,HPLC)测定七种不同产地酸枣仁中的皂苷A、B含量。结果:快速溶剂萃取最佳提取条件:两步提取,第一步提取的最佳条件是乙醇浓度60%、提取频率1次/min、提取压力100bar、提取温度100℃,第二步提取的最佳条件是乙醇浓度60%、提取频率5次/min、提取压力120bar、提取温度100℃,各地酸枣仁皂苷总含量大小依次为:河北白平>河北赞皇>山东>河北富平>辽宁>老挝>缅甸。结论:快速溶剂萃取法和HPLC-ELSD法测定酸枣仁皂苷回收率高、准确性好,产于河北、山东的酸枣仁中皂苷含量远高于其他地方的酸枣仁,可以作为开发酸枣仁保健品的优质原料使用。 

     

    Abstract: Objective: To develop the method of pretreatment and analysis for content determination of jujuboside A and B in Zizyphi Spinosi Semen ( ZSS) , to study the contents of jujuboside A and B in ZSS from different regions.Methods:Accelerated solvent extraction ( ASE) was used as method of extracting jujuboside A and B from ZSS and the conditions of ASE were optimized after parameters of ELSD were optimized and HPLC analysis method was tested, contents of jujuboside A and B in ZSS from seven different origins were determined.Results: The optimal ASE conditions: two-step extraction, optimum conditions of the first step were 60% ethanol, 1 times / min ( extraction frequency) , 100 bar ( extraction pressure) , 100℃ ( extraction temperature) , optimum conditions of the second step were 60% ethanol, 5 times / min, 120 bar, 100℃, the order of jujuboside contents in ZSS from different origins as follows: He Bei Baiping > He Bei Zanhuang >Shan Dong > He Bei Fuping > Liao Ning > Laos > Myanmar.Conclusion: HPLC- ELSD combined with ASE method is very suitable for determination of jujubosides A and B in ZSS with high recovery rate and good accuracy; ZSS produced in He Bei and Shan Dong in contents of jujubosides A and B is far higher than other ZSS, and these two ZSS can be used as high quality raw materials for researching and developing health care products.
