原生质体紫外诱变选育香菇耐高温菌株

Breeding thermo- tolerant strains of Lentinula edodes by UV mutagenesis

  • 摘要: 为了选育香菇耐高温新菌株,以香菇菌株沪农2号为出发菌株,紫外诱变处理其原生质体后,通过47℃处理2h初筛得到沪农2号耐高温诱变株24株。测定沪农2号及其所有诱变株在木屑培养基中的恒温长速、高温长速以及恢复长速,并进行高温出菇实验。对诱变株的生长速度及子实体性状进行综合分析发现,沪农2号的诱变株N14、N15、N21及N24的恢复长速快于出发菌株,表征子实体主要性状的单菇重量、菌盖厚度、菌盖直径也优于出发菌株,表明这4株诱变株的综合性状较好。 

     

    Abstract: In order to breed new thermo- tolerant strains of Lentinula edodes, the protoplasts of Hu Nong 2 ( original strain) were exposed to ultra- violet ray and the mutants were screened through mycelia recover ability after being treated at 47℃ for two hours.Totally 24 thermo- tolerant mutant strains of Hu Nong 2 were obtained.The growth rate of mycelia in sawdust medium during constant temperature process ( 25℃) , high temperature process ( 30℃) and recover temperature process ( 25℃) were determined. The fruiting bodies of the mutants were collected at 25℃.Comprehensive analysis of growth rate and biological characteristics of fruiting bodies showed that Hu Nong 2mutants N14, N15, N21 and N24 had faster growth rate during recover temperature process than Hu Nong 2 and at the same time these mutants showed better biological characteristics of fruiting bodies in single mushroom weight, cap thickness and cap diameter than Hu Nong 2.These results indicated that the four mutants of Hu Nong 2 had good comprehensive biological characteristics.
