超声波辅助提取牛睾丸中精蛋白工艺优化

Optimization of ultrasonic assistant protamine extraction processing parameters using response surface methodology in bovine testes

  • 摘要: 以牛睾丸为原料,确定超声波辅助法提取牛睾丸精蛋白的最优工艺条件。实验采用Box-Behnken设计模型确定最佳提取条件为:硫酸浓度5.5%、超声波功率190W、提取时间10min,牛睾丸精蛋白产率为4.19%,精蛋白含量为66.70μg/g,产率比硫酸提取法提高了33.87%,提取时间缩短至10min,蛋白质含量增加了6.47μg/g,选择95%的乙醇作为沉淀剂时,乙醇体积比1∶3时,所得精蛋白粗品的产率、水溶性和色泽均最佳。 

     

    Abstract: Experiments of extracting protamine with ultrasound assistant processing in bovine testes were designed using Box-Behnken response surface methodology. The optimal extraction processing parameters based on design expert were:5.5% sulfuric acid, 190 W of ultrasonic and extraction time was 10 min. Compared with the traditional extraction method, this study raised the protamine productivity up to 33.87%, decreased the extraction time to 10 min and increased the content of protamine with up to 6.47μg/g as well. In addition, while the optimal precipitating reagent was 95% ethanol, volumeration 1∶3 for testicular precipitation, protamine product yield, water soluble and color were the best.
