哈密瓜铜锌超氧化物歧化酶(Cu/Zn-SOD)基因的克隆及生物信息学分析

Cloning and bioinformatics analysis of Cu /Zn-SOD gene from cantaloupe

  • 摘要: 依据NCBI中已克隆的葫芦科植物黄瓜和西瓜铜锌超氧化物歧化酶(Cu/Zn-SOD)基因序列设计引物,以哈密瓜果肉总RNA为模板,通过RT-PCR扩增,最终获得一段403bp的哈密瓜Cu/Zn-SOD(HmCu/Zn-SOD)基因片段,运用Blast比对,系统进化树分析最终确定所克隆的基因片段为哈密瓜的Cu/Zn-SOD基因序列。运用蛋白质分析软件分析结果表明,克隆得到的哈密瓜Cu/Zn-SOD位于细胞质中,是非跨膜的非分泌性亲水蛋白,主要由无规则卷曲和β-折叠等蛋白质二级结构元件构成,同时与其他植物的胞质Cu/Zn-SOD的组成成分和理化性质具有高度的相似性。 

     

    Abstract: A superoxide dismutase gene was cloned by the Cu /Zn-superoxide dismutase gene ( Cu /Zn-SOD) from cucumber and watermelon has cloned in NCBI, which was named HmCu /Zn-SOD.The length cDNA of HmCu /Zn-SOD was 403bp.The characteristics were analyzed and predicted by the tools of bioinformatics.The results showed that HmCu/Zn-SOD was a hydrophilic, non-transmembrane and non-secretory protein.The main motif of Cu/ZnSOD in secondary structure was random coils and extended strands.Despite sequence variations, the structure and the composition of amino acid sequences of HmCu/Zn-SOD were highly similar to that of other green plants.
