链霉菌CA-1产磷脂酶D发酵条件的优化

Optimization of fermentation conditions of phospholipase D from Streptomyces sp.CA-1

  • 摘要: 以链霉菌CA-1菌为出发菌株,通过单因素实验和正交实验,对该菌株发酵产磷脂酶D的条件进行优化。结果表明最佳产酶条件为:葡萄糖20g/L、牛肉膏8.75g/L+鱼粉蛋白胨8.75g/L、吐温20 32.5g/L、氯化钙2.5g/L,接种量1.5%、培养基初始pH7.0、培养温度30℃、培养时间3d,在最佳发酵条件下,酶活达到1605.9U/L,是优化前的7.72倍。 

     

    Abstract: The Streptomyces sp.CA-1 as purpose strain, by single- factor experiment and orthogonal experiment, the fermentation conditions for phospholipase D production were optimized. Results showed that the optimal fermentation conditions for producing enzyme: glucose 20g /L, beef extract 8.75g /L + fishmeal peptone 8.75g /L, twain 20 32.5g /L, calcium chloride 2.5g /L, inoculums 1.5%, medium initial pH 7.0, culture temperature 30℃, incubation time 3d.Under the best fermentation conditions, the enzyme activity could reach 1605.9U /L, which was7.72 times than that of unoptimized conditions.
