人参脂溶性成分提取工艺优化及其GCMS分析

Optimization of extraction process for lipid-soluble components in ginseng and its GC-MS analysis

  • 摘要: 以5年人工种植人参为原料,采用超临界CO2萃取法萃取人参脂溶性成分,通过正交实验对萃取工艺条件进行优化,并用气相色谱-质谱技术(GC-MS)对萃取物的化学成分进行定性定量分析,结果表明:在萃取压力25MPa、萃取温度40℃、萃取时间3.5h、原料与夹带剂之比(g/mL)为1∶0.6时,人参脂溶性成分的萃取率最高,达85.38%±3.16%。从超临界CO2萃取法提取的人参脂溶性成分中鉴定出28种化合物,以脂肪酸类、酯类、甾醇类、倍半萜类、烯烃类化合物为主,其中相对含量较高的3种组分是亚油酸(43.12%)、顺式-6-十八碳烯酸(21.65%)和棕榈酸(11.86%)。 

     

    Abstract: With five- year cultivated ginseng as raw material, the lipid- soluble components in ginseng were extracted using supercritical CO2extraction technique.The conditions of extraction process were optimized through orthogonal design test and chemical compositions of extract were identified and quantified by gas chromatography-mass spectrometry ( GC- MS) .The results showed that the highest extract yield of ginseng lipid- soluble components was 85.38% ± 3.16%, with extraction pressure at 25MPa, extraction temperature at 40℃, extraction time at 3.5h and a material to entrainer ratio ( g /mL) at 1 ∶ 0.6.A total of 28 compounds were indentified from the ginseng lipid- soluble components extracted by supercritical CO2, among which fatty acids, esters, sterols, sesquiterpenes and alkene were the principal constituents.Three components including linoleic acid ( 43.12%) , 6-octadecenoic acid ( 21.65%) and hexadecenoic acid ( 11.86%) were present in higher relative amount.
