多粘类芽孢杆菌PS04胞外多糖分子结构研究

Study on the molecular structure of polysaccharide produced by PS04 strain

  • 摘要: 采用丙烯葡聚糖凝胶S-200 HR对PS04多糖进行过滤层析,得到单一组分峰,且通过凝胶渗透色谱法分析测定,可知该多糖重均分子量为4009ku,峰位分子量为3174ku,分布宽度指数为1.348;结合红外光谱、高效液相色谱及核磁共振分析,确定该多糖由果糖与葡萄糖以7∶1的比例组成,在多糖结构中,果糖残基以呋喃环构型存在,且以β-2,6、β-1,2键连接,因PS04多糖与果聚糖最为接近,可初步判断该多糖是一种果聚糖。 

     

    Abstract: A single component peak was found in PS04 polysaccharide purity analysis using S-200 HR filtration of chromatography. The weight mean molecular weight of the PS04 polysaccharides examined by GPC was4009ku, the peak positions molecular weight was 3174ku, and distribution width index was 1.348.The structure of PS04 polysaccharide was identified by infrared spectral, HPLC, and NMR. The results showed that PS04polysaccharide was a typical polysaccharide compounds and the main composition was fructose and glucose (proportion 7∶1) . NMR analysis demonstrated that there were furan nucleus and the chain was linked by β-2, 6and β-1, 2 linkage, so PS04 polysaccharide was similar to levan.
