嗜酸乳杆菌NCFM表层蛋白的提取及其与菌体酸和胆盐耐受及粘附性能的关系

Extraction of Surface Layer Protein from Lactobacillus acidophilus NCFM and the relationship among SLP and strain acid, bile resistance and cell adhesion

  • 摘要: 对嗜酸乳杆菌NCFM表层蛋白(SLP)的氯化锂(LiCl)提取方法进行了优化,通过SDS-PAGE验证,确定了效率最高的提取条件,即用5mol/L LiCl在35℃处理菌体20min,且提取SLP后,菌体不会失活,8h后重新生成的SLP达到峰值。利用SLP能够再生的性质,分析了SLP与菌体耐酸耐胆盐性能,以及菌体对肠上皮细胞HT-29粘附能力的关系。实验结果显示:随着SLP的再生,去除SLP的菌体酸存活率由0h的52.5%上升到8h后的85.9%,胆盐存活率由0h的43.4%上升到8h后的81.8%,菌体对HT-29细胞的粘附指数从0h的0.33上升到8h后的18.54。SLP再生过程中菌体对酸和胆盐的耐受力和对HT-29细胞的粘附力均有恢复,因此说明SLP的完整对嗜酸乳杆菌NCFM在消化中存活力有重要影响。 

     

    Abstract: Extraction method of Surface Layer Protein from Lactobacillus acidophilus NCFM with LiCl was optimized.SDS-PAGE was employed to validate the extraction performance, and the optimized condition was that the strain culture was treated with 5mol /L LiCl for 20min at 35℃.After SLP extraction, the activity of strain was remained and the surface protein could reform to the maximum in 8h.The property of SLP reformation was employed to analyze the relationship among SLP and strain acid / bile resistance, and the adhesion of strain to HT-29 cell.The results showed that, after 8h SLP reformation, the acid challenge survival rates of SLP removed strains enhanced from52.5% to 85.9%, and bile challenge survival rates of SLP removed strains enhanced from 43.4% to 81.8%.And the adhesion index to HT- 29 increased from 0.33 to 18.54 after 8h SLP reformation. The indicators related to strain survival in GI track were recovered as the reformation of SLP.In conclusion, the integrity of SLP was important to the survival of Lactobacillus acidophilus NCFM in GI track.
