响应面法优化红缘拟层孔菌胞外多糖发酵培养基

Optimization of culture medium of Fomitopsis pinicola for exopolysaccharide production by response surface methodology

  • 摘要: 为提高红缘拟层孔菌(Fomitopsis pinicola)胞外多糖(EPS)产量,采用响应面法对其发酵培养基进行优化。在单因素实验基础上,利用中心组合设计原理,以蔗糖、酵母粉、MgSO4·7H2O浓度为实验因素,以多糖产量为响应值,采用3因素5水平响应面分析方法建立数学模型。结果显示,红缘拟层孔菌产EPS的最佳培养基为:蔗糖186g/L、酵母粉7g/L,MgSO4·7H2O 27g/L,KH2PO42g/L。在此条件下,EPS产量预测值为4.02g/L,验证值为(3.92±0.18)g/L。采用响应面法对红缘拟层孔菌EPS发酵培养基进行优化,方法可行。 

     

    Abstract: Response surface methodology was employed to optimize the culture medium for exopolysaccharide production from Fomitopsis pinicola. Exopolysaccharide yield was investigated with respect to the concentration of sucrose, yeast extract and MgSO4 ·7H2 O based on these results of single factor experiment. A quadratic regression model was established on a three-variable, five-level center composite design. The optimum concentration of sucrose, yeast extract and MgSO4 ·7H2 O were 186g/L, 7g/L and 27g/L, resepectively. Under these conditions, the predicted value of EPS yield was 4.02g/L, while the actual value was (3.92±0.18) g/L. So response surface methodology was feasible for optimization of fermentation medium of Fomitopsis pinicola for EPS production.
