两步酶法提取玉米胚芽蛋白的工艺研究

Study on extraction of corn germ protein by two-step enzymatic method

  • 摘要: 为确定更为温和、高效的玉米胚芽蛋白提取工艺,本实验采用纤维素酶和碱性蛋白酶对玉米胚芽进行两步酶法处理。通过单因素考察和正交实验设计对提取工艺参数进行优化。结果表明,纤维素酶酶解的最佳工艺为:pH4.5、酶解时间2.5h、加酶量0.3%(W/V);碱性蛋白酶酶解的最佳工艺为:pH9.0、酶解时间3h、加酶量4%(V/V)。在此条件下,玉米胚芽蛋白的提取率可达到83.7%±1.2%。 

     

    Abstract: In order to investigate gentler and more efficient corn germ protein extraction method, the corn germ protein was extracted from corn germ by the two-step enzymatic method ( cellulase and alkaline protease) .Single factor experiments and orthogonal experiments were performed to optimize the extraction processing parameters. The results showed that, the optimum conditions for cellulase processing were as follows: pH4.5, reaction time 2.5h and enzyme dosage 0.3% ( W/V) .The optimum conditions of alkaline protease processing were as follows: pH 9.0, reaction time 3h and enzyme dosage 4% ( V/V) . Under the optimum conditions, the extraction rate of corn germ protein was 83.7% ± 1.2%.
