响应面法优化纤维素酶提取独活蛇床子素工艺的研究

Optimization of cellulase-assisted extraction of osthole from root of Radix Angelica Pubescens using response surface methodology

  • 摘要: 采用响应面分析法(RSM)优化纤维素酶提取蛇床子素的工艺条件。在单因素实验的基础上,选取酶解pH、纤维素酶量、酶解时间、酶解温度作为影响因子,以蛇床子素得率为响应值,利用Box-Benhnken中心组合设计原理,研究各自变量及其交互作用对得率的影响,模拟得到二次多项式回归方程的预测模型,确定最佳提取条件为:酶解pH为5.0,酶解时间60min,酶解温度50℃,纤维素酶量3%。在此优化工艺条件下,蛇床子素的得率为0.926%。 

     

    Abstract: Response surface methodology ( RSM) was applied to optimize the extraction conditions of osthole.Based on single factor experiment, pH, the amount of cellulase enzymolysis temperature and time were chosen as influencing factors, the yield of osthole was selected as response value.The mathematical model was established by Box- Behnken central composite design.The results showed that the optimal extraction conditions were as follows: pH5.0, the amount of cellulase 3%, enzymolysis temperature 50℃, enzymolysis time 60min.In this optimal process condition, the yield of osthole was 0.926%.
