液相色谱-串联质谱法测定水产品中β-内酰胺类抗生素的残留

Determination of 6 β-lactam antibiotic residues in aquatic product by high performance liquid chromatography-tandem mass spectrometry

  • 摘要: 建立了同时测定水产品中6种β-内酰胺类抗生素药物残留的高效液相色谱-串联质谱(HPLC-MS/MS)检测方法。样品经乙腈-水(15∶2,v∶v)混合液提取,HLB固相萃取柱净化。以乙腈-0.01mol/L醋酸铵溶液(pH4.5)作为流动相,C18色谱柱进行分离,电喷雾正离子多反应监测模式进行检测。方法检测限为0.2~2.0μg/kg,加标回收率为80.8%~92.9%,相对标准偏差为2.6%~9.1%(n=9)。该方法准确、灵敏、简便,适用于水产品中6种β-内酰胺类抗生素残留的测定。 

     

    Abstract: A high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) method was established for simultaneous determination of 6 β-lactam antibiotic residues in aquatic product.Samples were extracted with acetonitrile - water ( 15 ∶ 2 , v ∶ v ) and purified by Oasis HLB solid phase extraction column . Chromatographic separation was carried out on a C 18 column, the mobile phase was acetonitrile -0.01mol/L ammonium acetate solution ( pH4.5 ) with gradient elution , then residues were detected by MS / MS in the ESI (+) -MRM mode. Limits of detection for the method were 0.2~2μg/kg, recoveries ranged from 80.8%~92.9%, and relative standard deviations (RSD) were in the range of 2.6%~9.1% (n=9) . The method was accurate, sensitive, simple and suitable for the determination of six β-lactam antibiotic residues in aquatic product.
