亚致死热损伤和复苏状态的副溶血性弧菌特性

Characteristics of the sublethal injured and resuscitative Vibrio parahaemolyticus

  • 摘要: 目的:研究副溶血性弧菌亚致死损伤和复苏后的生长特性和蛋白质、核酸损伤情况,并观察损伤后细菌的形态特征,检测损伤和复苏菌株的生理生化特性和致病基因。方法:对副溶血性弧菌VpBJ1.1616和VpBJ1.1997的亚致死热损伤和复苏后菌株的生长曲线进行测定,菌液离心后上清液测定OD260nm和OD280nm用以检测核酸和蛋白质流失情况,扫描电镜观察各状态下细菌的形态特征,并对正常状态、亚致死损伤状态和复苏后的副溶血性弧菌进行了生理生化和致病基因的检测。结果:损伤后的菌株迟滞期长、核酸蛋白质流失与正常细菌有显著差异(p<0.05),而复苏与正常菌株生长情况相近,且VpBJ1.1616的核酸蛋白质物质流失与正常细菌没有明显差异(p>0.05)。损伤的副溶血性弧菌表面有明显地破损,某些生理生化特性发生了不可逆的改变,但是致病基因与正常菌株基本相同。结论:通过对损伤和复苏后各种特性的检测,为研究副溶血性弧菌亚致死损伤状态和食品安全控制提供理论依据。 

     

    Abstract: Objective:In order to research the characteristics of the sublethal injured and the resuscitative Vibrio parahaemolyticus, the morhphological characteristics and the virulence genes were investigated.Methods:The growth curses of sublethal injured and resuscitative Vibrio parahaemolyticus BJ1.1616 and BJ1.1997 were made, and then the cellular materials leakage of Vp was determined.The morphological characteristics of the sublethal injured and the resuscitative strains were observed by using SEM, and the physiological and biochemical characteristics and the virulence genes were detected.Results:The injured strains showed an extended lag phase, but the growth curses of the resuscitative strains were similar with the normal strains.The supernatant of the cell suspension after thermal treatment showed a significantly (p<0.05) higher absorbance at 260 and 280nm when compared with that of the control cells, but the resuscitative cell did not differ significantly (p>0.05) .There was no difference between VpBJ1.1616 and normal bacteria in 260nm and 280nm absorbing materials.Compared with the normal strains, the morphological characteristics of sublethal injured and resuscitative strains and some of the physiological and biochemical characteristics changed significantly and irreversible, but the virulence genes remained.Conclusion:These data of sublethal injury and resuscitative conditions can provided a basis on the mechanism study, as well as the detection of sublethal injury Vp.
