阿魏酸酯酶活性的分光光度法测定及影响因素研究

Study on the determination of ferulic acid esterase enzyme activity with spectrophotometer method and its impact factors

  • 摘要: 分别以去淀粉麦麸和对硝基苯酚阿魏酸酯为底物,研究了紫外-可见光分光光度法测定阿魏酸酯酶酶活的方法,通过测定阿魏酸的释放量和对硝基苯酚的释放量来计算阿魏酸酯酶的活力,并研究了酶活测定的影响因素。同时,对去淀粉麦麸分光光度法测定酶活进行了方法学验证,得到平均回收率达99.86%,RSD值为0.65%。并将此方法与对硝基苯酚阿魏酸酯分光光度计法及HPLC法测定阿魏酸酯酶酶活进行了比较,结果表明去淀粉麦麸分光光度计法测定阿魏酸酯酶酶活具有稳定性好,相对偏差较小的优点且简便可行。 

     

    Abstract: The UV-Vis spectrophotometer methods to determination the ferulic acid esterase enzyme activity was studied using destarchedwheat bran (DSWB) and 4-nitrophenyl ferulate as the substrates respectively.The enzyme activity was calculated by measuring the quantities of ferulic acid released and the quantities of p-nitrophenol released.The factors influencing the enzyme activity determination were also investigated.The method for the determination the ferulic acid esterase enzyme activity using DWB substrate was validated with the average recovery of 99.86% and RSD was 0.65% . It was compared with the method of 4-nitrophenyl ferulate spectrophotometer and the method of HPLC.The results showed that the DSWB spectrophotometer method had advantages of a good stability, smaller relative deviation, simple and feasible.
