酶解星虫水溶性蛋白制备降血压肽的工艺研究

  • 摘要: 以可口革囊星虫水溶性蛋白为原料,利用胃蛋白酶、碱性蛋白酶、胰蛋白酶、木瓜蛋白酶和中性蛋白酶水解星虫蛋白制备降血压肽;并以酶解产物对血管紧张素转化酶(ACE)的抑制率为指标,对酶解条件进行优化,确定了星虫蛋白酶解的最佳条件为酶解时间4h、酶解pH为1.8、酶添加量为0.3%(w/v),温度37℃,所得酶解产物的IC50为1.85mg/mL。同时对酶解产物的体内降血压活性进行了动物实验,研究表明,星虫蛋白降血压肽在给药后1h,收缩压(SBP)开始下降,一直持续8h,SBP平均降低了约17mmHg。 

     

    Abstract: Phascolosoma esculenta water-soluble protein hydrolyzed by pepsin, alkali protease, trypsin, neutral protease, and papain were studied. The hydrolysis condition of pepsin was optimized, with the ACE inhibiting rate as the major indicator, to:pH1.8, 37℃ , E/S 0.3% (w/v) and 4h. Under the optimized condition of pepsin hydrolysis, the IC 50 of hydrolysate was 1.85mg/mL. The activity of ACE inhibitor in vivo was further investigated with spontaneously hypertensive rats (SHR) and results showed that blood pressure start to decrease significantly after 1h of oral administration and the average value of SBP reduced 17mmHg in 8h.
