超滤分离纯化菠萝蛋白酶的研究

  • 摘要: 采用超滤分离纯化菠萝蛋白酶,考察了超滤操作压力、温度、浓缩倍数等因素对膜通量和截留率这两个超滤的重要指标的影响,结果在理想状态(压力(0.184±0.007)MPa、温度(10±0.5)℃、浓缩倍数16倍)时,菠萝蛋白酶的纯化倍数达到89.4倍,酶活回收率相应达到79.3%,比TiO2多孔陶瓷柱层析纯化倍数多81.4倍,且回收率提高了31.3%。超滤法大大减少实验使用场地和设备使用量,酶活回收率好、所得比酶活率高,起到高度纯化的作用,而且此法操作简单、处理果汁量大,反映超滤法较适合生产高纯度菠萝蛋白酶。 

     

    Abstract: Ultra-filtration was used to isolate and purify bromelain , and operating pressure, temperature, concentration and other factors were investigated on the impact of two important indicators for the flux of membrane and rejection ratio of ultra-filtration. The results indicated that the purification ratio of bromelain reached 89.4 times, and recovery ratio of enzyme activity was related up to 79.3%, which was more 81.4 times than the TiO2 porous ceramic column. Furthermore, the recovery ratio was increased by 31.3% in the desired state (pressure (0.184±0.007) MPa, temperature (10±0.5) ℃, concentration ratio 16) . Test venues and apparatus used amount were significantly reduced when isolation and purification of bromelain was used of the ultra-filtration method. Meanwhile, the method was played a role in a highly purified effect because good recovery ratio of enzyme activity and high ralative activity were obtained. In addition, the procedure was convenient in operation, small in volume, large in capacity, reflecting that the ultrafiltration method was relatively suitable for the production of high purity bromelain.
