粪肠球菌SK23.001产瓜氨酸发酵条件的优化研究

Study on optimization on fermentation conditions of producing citrulline by Enterococcus faecalis SK23.001

  • 摘要: 从西瓜大棚的土壤中筛出一株可以发酵生产瓜氨酸的菌株SK23.001,经鉴定为粪肠球菌(Enterococcus faecalis)。为进一步提高瓜氨酸的产量,对菌株SK23.001的发酵条件进行优化。确定了最佳接种菌龄为12h,发酵时间为16h。单因素实验和正交优化实验确定了最佳的发酵培养条件:碳源为蔗糖0.5%,氮源为大豆蛋白胨和NH4Cl,接种量3%,装液量50mL/250mL,起始pH6.0,温度37℃;100g/L的L-Arg转化反应6h,经HPLC检测产量可达83.06g/L。 

     

    Abstract: A novel strain named SK23.001 was isolated from soil in Watermelon canopy and it was identified as Enterococcus faecalis. In order to enlarge the yield of citrulline, the fermentation of Enterococcus faecalis SK23.001 were optimalized. The optimal vaccination age and culture time was 12h and 16h, respectively. The optimum fermentation conditions by single -factor experiment and orthogonal experiment were determined:carbon source sucrose (0.5%) , nitrogen source soya peptone and ammonium chloride, inoculation volume 3%, liquid medium volume 50mL/250mL, pH6.0, temperature 37℃ . The product was analyzed by HPLC and the 83.06g/L of citrulline transformed from 100g/L arginine could be attained after 6h.
