陈皮中橙皮苷的提取与纯化工艺研究

Study on the extraction and purification of hesperidin from Pericarpium Citri Reticulate

  • 摘要: 主要研究了乙醇热回流法从陈皮条中提取橙皮苷的最优浸提条件,并对乙醇、蒸馏水洗涤纯化橙皮苷粗品的工艺条件进行了探索。结果表明,陈皮条中橙皮苷的最佳提取工艺条件为:乙醇浓度75%(v/v),料液比1∶35(w/v),提取温度85℃,提取时间2.5h。建立了简便、有效的粗提物洗涤纯化工艺:0.1g橙皮苷粗品用10mL95%乙醇搅拌洗涤20min,重复2次,相同条件下再用蒸馏水洗涤1次。采用该方法提取及纯化的橙皮苷经高效液相色谱检测,提取得率为3.28%±0.1%,纯度达到93.1%±1.1%,回收率为70.7%±2.2%。 

     

    Abstract: An effective extraction and purification technology of hesperidin from Pericarpium Citri Reticulate strip was established:hesperidin was extracted by ethanol refluxing and the crude extract was purified via ethanol washing followed by distilled water washing. The most optimal extraction conditions were as follows:75% (v/v) ethanol refluxing at 85℃ for 2.5h, the ratio of Pericarpium Citri Reticulate to extracting liquid was 1∶35 (w/v) . The most optimal purifying conditions were that 1.0g crude extract was washed with 10mL 95% ethanol for 20min, repeated 2 times, then washed with distilled water once. HPLC analysis showed that the yield rate of hesperidin was 3.28% ±0.1% raw material, the purity of the final products and the recovery rate were 93.1% ±1.1% and 70.7%±2.2%, respectively.
