亚甲基蓝对肠出血性大肠杆菌O157的光动力杀菌技术研究

Anti-microbial photodynamic technology of methylene blue against Enterohaemorrhagic Escherichia coli O157

  • 摘要: 研究了亚甲基蓝对革兰氏阴性菌肠出血性大肠杆菌O157的光动力杀伤作用的条件参数。采用单因素实验比较了亚甲基蓝浓度、光照时间、孵育时间对O157菌落数的影响。在此基础上运用Box-Behnken的中心组合实验设计,优化了该技术的条件参数,建立了各因子与O157菌落数关系的数学回归模型,确定了最佳的反应条件,即亚甲基蓝浓度为24.08mg/L,光照时间为27.66min,孵育时间为19.00min时,O157的菌落数对数为1.03,与预测的理论值1.00极为接近。 

     

    Abstract: Condition parameters for anti-microbial photodynamic technology (APDT) of methylene blue (MB) against Enterohaemorrhagic Escherichia coli (EHEC) O157 was investigated. The influence of concentration of MB, illumination time and incubation time on colony-forming unit (CFU) were compared by single factor experiment. The mathematical model describing the relationship between the CFU of O157 and each factor was developed in order to optimize the condition parameters by Box-Behnken center combination experimental design. The optimum values of the above conditions were obtained as:illumination for 27. 66min and incubation for 19. 00min under the presence of 24. 08mg/L MB. Under such conditions, the population of O157 was down to 1. 03logCFU, close to the theoretical prediction value of 1. 00logCFU.
