甜玉米穗轴提取水溶性木聚糖的研究

Extraction of water-soluble xylan from sweet corn cobs

  • 摘要: 旨在寻找一种甜玉米穗轴中水溶性木聚糖的提取和纯化方法。以甜玉米穗轴为原料,采用酸预处理高温蒸煮法提取木聚糖,正交实验后获得提取方法的最佳条件,并验证其提取率。木聚糖液经脱色、脱蛋白处理后,使用CaptoQ阴离子交换树脂对木聚糖液进行纯化。结果如下:3g/L的H2SO4浸泡12h后,料液比为1∶10,蒸煮温度为150℃,蒸煮时间为60min,提取率为27.32%。柱层析纯化后,木聚糖提取液的收率为51.8%,冻干粉木聚糖含量为82.7%。本文对甜玉米穗轴的木聚糖提取和纯化工艺进行了可行性研究和数据支持。 

     

    Abstract: The aim was to establish a method of extracting and purifying Water-Soluble xylan from sweet corn cobs. Xylan was extracted from sweet corn cobs through stewing method and the optimized conditions of the extraction method were confirmed by the orthogonal experiment. The extraction efficiency was determined subsequently. Simultaneously xylan from sweet corn cobs was purified by Capto Q after disposing of decoloration and removing protein. The results were shown as follows:sweet corn cobs were soaked by 3g/L H2SO4 after 12h, the ratio of solid to liquid 1∶ 10 (g∶ mL) , the temperature and time of stewing were 150℃ and 60min, respectively. Using column chromatography, the dry powder content and yield of xylan were 82. 7% and 51. 8%, respectively. The optimum extraction ratio of this method was 27. 32%. In this study, the water-soluble xylan of sweet corn cobs was extracted and purified, the supporting of experiment data were provided for the future xylan industrialization.
