Study on the effect of different pretreatment condition to the determination of auricularia auricula dietary fiber

  • 摘要: 针对黑木耳膳食纤维的特性,采用AOAC推荐的酶-重量法和酶-化学法均没有考虑非蛋白氮,不能准确地测定黑木耳膳食纤维的含量。本文通过黑木耳中非蛋白氮的测定,对比AOAC Official Method 991.43法及该方法校正后对测定黑木耳膳食纤维的差异。首先以甲壳素为标样,测定其中的氨基糖回收率,确定微波消解样品成游离氨基糖的最佳条件为盐酸浓度8mol/L、盐酸用量8mL、消解第一阶段功率60W、消解60s,第二阶段功率100W、消解120s;在此条件下处理黑木耳样品后,参照AOAC 991.43法测定黑木耳膳食纤维含量时会造成3.18%的偏差。故测定含几丁质的食用菌类物质的膳食纤维时需对AOAC方法进行校正。 

     

    Abstract: For the problem of the nitrogen element in the aminoglycoside molecule in the cell wall of Auricularia auricular, it was not accurate to determine the contents of total dietary fiber of auricularia auricula samples by enzyme-weighted method and enzyme-chemical method by AOAC directly, without considering the non-protein nitrogen. The non-protein nitrogen in the auricularia auricula samples were determined in this paper, and the difference on the determination auricularia auricula dietary fiber was compared between AOAC official method 991. 43 and its correction method. Firstly, the optimum condition to determine the recovery of aminoglycoside in chitin was gained, first stage was 60W and 60s, and second stage was 100W and 120s, upon the condition of 8mol/L and 8mL HCl at microwave digestion method. The result showed that there would be 3. 18% deviation with AOAC official method 991. 43 directly, and the correction to AOAC official method was necessary at the determination of edible fungi dietary fiber with chitin.
