大孔树脂吸附分离蚕蛹ACE抑制肽的研究

  • 摘要: 为了找出大孔树脂纯化蚕蛹ACE抑制肽的方法。比较了5种大孔树脂在纯化蚕蛹ACE抑制肽时的吸附率和解吸率,得出DA201-C分离蚕蛹ACE抑制肽的效果最好。再进一步考察上样流速、上样浓度、分级洗脱等对DA201-C大孔树脂吸附及分离效果的影响。最终结果为75%乙醇洗脱下的多肽的ACE抑制率最高,其IC50为0.632mg/mL,并且发现疏水性与ACE抑制率呈正相关。 

     

    Abstract: The purpose of this experiment was to find out the method of macroporous resin purifying pupa ACE inhibitory peptides. Through comparing the absorption rate and desorption rate of 5 kinds of macroporous resins in purifying pupa ACE inhibitory peptides, it was the result that DA201-C had the best effect in purifying pupa ACE inhibitory peptides.Further research of the impact of sample's flow, concentration and graded elution on macroporous resin's separating effect.The final conclusions were that when the ethanol elution concentration was 75%, the eluent had the highest ACE inhibition rate, whose IC50 was 0.632mg/mL and it was found that hydrophobicity and ACE inhibition rate were positively correlated.
